Council currently has in place an interim ban of the chemical.
Cr Horton's attempt to implement the permanent ban came after council received a report updating the council on an interim ban.
Councillors had been asked to receive and note the report which mayor Brian Ingram noted told the council "absolutely nothing and makes no decision".
"It doesn't give councillors any guidance on where we should or shouldn't go," Cr Ingram said of the report.
Glyphosate is a broad-spectrum herbicide which has been commonly used for weed control over many years.
There has recently been a number of cases in the United States which have found that Glyphosate has been the main contributory cause to certain cancers.
Attempting to ban its use by council staff Cr Horton said the World Health Organisation had proven proved that Gyphosate causes cancer and kills people.
"We should ban the use of it," he said.
"Since I've lived in Harden I have never seen so many people die of cancer.
"From where I came from you hardly ever heard of it.
"It's a great weed killer but it kills people and I think we should ban the use of it, let's get rid of it.
"I'd like to move we ban completely the use of Glyphosate from use by Hilltops council. It's a great weed killer but it should be banned," Cr Horton said.
In support of Cr Horton, Cr Margaret Roles said "coming from a farming family Glyphosate is used by is but increasingly we are looking at other means and chemicals".
"To say that other chemicals are carcinogens is fine but is no reason why we don't work to eliminate as much as we can, if by cutting down use of Glyphosate in Hilltops we are protecting those working and who live within Hilltops than I support that motion," Cr Roles said.
Cr Horton and Cr Roles were the only two councillors to support Cr Horton's move to make the interim ban of Glyphosate a permanent ban.
Speaking against receiving and noting the report and moving a new motion Cr Flanery pointed out there are a number of known carcinogens which aren't banned, including diesel fuel and smoking.
"This is the only effective complete weed killer in a council where we are reliant on agriculture.
"This is an extreme, all of the components are naturally occurring chemicals in the environment.
"I think there are adequate protections we can put in place to enable roadside spraying to control noxious weeds," he said.
Cr Flanery called for a further report to come back to the council "detailing what, if any adequate WHS safety measures can be put in place to enable the continued use of Glyphosate to control noxious weeds on roadsides to protect the agricultural industry".
Councillors voted to support Cr Flanery's motion, with the exception of Cr Horton, Cr Rita O'Connor and Cr Roles.
